We have developed the unique ToxTracker®, ReproTracker® and ToxProfiler assays. These high throughput stem cell-based reporter assays allow for reliable carcinogenicity, developmental and mechanistic toxicity hazard identification. Our assays provide mechanistic insight into the reactive properties of chemicals and can be particularly useful in Adverse Outcome Pathway (AOP) approaches for chemical safety assessment.

  • ToxTracker suite
    The ToxTracker suite contains a unique collection of in vitro genotoxicity assays that can reliably identify genotoxic compounds and can provide detailed insight into their mode-of-action....

    ToxTracker consists of a collection of 6 unique fluorescent reporter cell lines that are combined in a single assay. These reporters signal the activation of various cellular stress responses that indicate the induction of DNA damage, oxidative stress, and protein damage. The extended assay, ToxTracker ACEcombines these different fluorescent reporters with an extensive cell cycle analysis and polyploidy analysis to discriminate between genotoxins with a clastogenic or an aneugenic mode-of-action. Aneugens can be further characterised in TubulinTracker to support an Adverse Outcome Pathway (AOP) approach. Genotoxic effects that are indirectly caused by high levels of oxidative stress can be readily identified in ToxTracker AO. The ToxTracker suite is particularly useful for AOP and read-across approaches. All assays can be useful as stand-alone or as follow-up tests of ToxTracker.

    ToxTracker is typically used for early genotoxicity screening and as a follow-up test for mode-of-action assessment. ToxTracker requires small amounts of test material, has a short turn-around time, and has an excellent predictivity for the Ames mutation test, as well as, the in vitro and in vivo micronucleus assay. The mode-of-action information from the ToxTracker suite can be highly valuable during compound development or in a weight-of-evidence approach for regulatory application.  

  • ReproTracker
    ReproTracker is a state-of-the-art in vitro assay to visualise the key events during early embryonic development that allows us to reliably assess the potential developmental toxicity hazards of new drugs and chemicals....

  • ReproTracker® is a human induced pluripotent stem cells (hiPSCs)-based assay, which combines the assessment of functional/morphological changes with expression patterns of tissue-specific biomarkers to detect possible developmental toxicity responses of chemicals.

  • ToxProfiler is a human cell assay that contains seven fluorescent reporter genes to visualise the major cellular stress response pathways. Based on the reporter gene activation patterns, ToxProfiler provides a detailed toxicological fingerprint for each test compound. Quantitative dose response modelling of the toxicological responses and point-of-departure calculations allow for potency ranking of compounds, identification of their primary mode-of-action, and read-across approaches for chemicals. ToxProfiler is particularly applicable for early safety testing, adverse outcome pathway (AOP) and weight-of- evidence (WoE) methods.
